Summary

Human chorionic gonadotropin (hCG) is a glycoprotein hormone produced by the developing placenta shortly after fertilization. In a normal pregnancy, hCG can be detected in urine and serum as early as 7 to 10 days after conception. The levels of hCG rise rapidly, often exceeding 100mIU/mL by the time of the first missed menstrual period and peaking at 100,000-200,000mIU/mL around 10-12 weeks into pregnancy. The presence and rapid increase of hCG make it an excellent marker for early pregnancy detection.

Test Sensitivity

The hCG Pregnancy Rapid Test Cassette detects hCG in urine at a sensitivity level of 25mIU/mL. It employs a combination of monoclonal and polyclonal antibodies to specifically identify elevated hCG levels, ensuring no cross-reactivity with structurally related glycoprotein hormones such as hFSH, hLH, and hTSH at high physiological levels.

Principle of the Test

This rapid chromatographic immunoassay utilizes two lines to indicate the results:

  • Test Line (T): Comprises monoclonal hCG antibodies that detect elevated hCG levels in the urine.
  • Control Line (C): Contains goat polyclonal antibodies and colloidal gold particles.

When a urine specimen is added to the test cassette, it migrates via capillary action along the membrane and reacts with the colored conjugate. A positive result is indicated by a colored line in the test line region, formed by the reaction of the antibody-hCG-colored conjugate with the hCG in the specimen. Absence of this line indicates a negative result. The control line appears to confirm that a proper volume of specimen has been added and that the membrane wicking process has occurred.

Usage Instructions

  1. Preparation: Ensure all components and specimens are at room temperature.
  2. Testing Procedure: Add the urine specimen to the specimen well of the test cassette.
  3. Results Interpretation:
    • Positive: Two colored lines appear (one in the test region and one in the control region).
    • Negative: Only one colored line appears in the control region.
    • Invalid: No lines or only a test line appears, indicating the test should be repeated with a new cassette.
